DRAGON-Data: a platform and protocol for integrating genomic and phenotypic data across large psychiatric cohorts

Background
Current psychiatric diagnoses, although heritable, have not been clearly mapped onto distinct underlying pathogenic processes. The same symptoms often occur in multiple disorders, and a substantial proportion of both genetic and environmental risk factors are shared across disorders. However, the relationship between shared symptoms and shared genetic liability is still poorly understood.
AimsWell-characterised, cross-disorder samples are needed to investigate this matter, but few currently exist. Our aim is to develop procedures to purposely curate and aggregate genotypic and phenotypic data in psychiatric research.
MethodAs part of the Cardiff MRC Mental Health Data Pathfinder initiative, we have curated and harmonised phenotypic and genetic information from 15 studies to create a new data repository, DRAGON-Data. To date, DRAGON-Data includes over 45 000 individuals: adults and children with neurodevelopmental or psychiatric diagnoses, affected probands within collected families and individuals who carry a known neurodevelopmental risk copy number variant.
ResultsWe have processed the available phenotype information to derive core variables that can be reliably analysed across groups. In addition, all data-sets with genotype information have undergone rigorous quality control, imputation, copy number variant calling and polygenic score generation.
ConclusionsDRAGON-Data combines genetic and non-genetic information, and is available as a resource for research across traditional psychiatric diagnostic categories. Algorithms and pipelines used for data harmonisation are currently publicly available for the scientific community, and an appropriate data-sharing protocol will be developed as part of ongoing projects (DATAMIND) in partnership with Health Data Research UK.
Faculty development in the age of competency-based medical education: A needs assessment of Canadian emergency medicine faculty and senior trainees

Objectives
The Royal College of Physicians and Surgeons of Canada (RCPSC) emergency medicine (EM) programs transitioned to the Competence by Design training framework in July 2018. Prior to this transition, a nation-wide survey was conducted to gain a better understanding of EM faculty and senior resident attitudes towards the implementation of this new program of assessment.
MethodsA multi-site, cross-sectional needs assessment survey was conducted. We aimed to document perceptions about competency-based medical education, attitudes towards implementation, perceived/prompted/unperceived faculty development needs. EM faculty and senior residents were nominated by program directors across RCPSC EM programs. Simple descriptive statistics were used to analyse the data.
ResultsBetween February and April 2018, 47 participants completed the survey (58.8% response rate). Most respondents (89.4%) thought learners should receive feedback during every shift; 55.3% felt that they provided adequate feedback. Many respondents (78.7%) felt that the ED would allow for direct observation, and most (91.5%) participants were confident that they could incorporate workplace-based assessments (WBAs). Although a fair number of respondents (44.7%) felt that Competence by Design would not impact patient care, some (17.0%) were worried that it may negatively impact it. Perceived faculty development priorities included feedback delivery, completing WBAs, and resident promotion decisions.
ConclusionsRCPSC EM faculty have positive attitudes towards competency-based medical education-relevant concepts such as feedback and opportunities for direct observation via WBAs. Perceived threats to Competence by Design implementation included concerns that patient care and trainee education might be negatively impacted. Faculty development should concentrate on further developing supervisors’ teaching skills, focusing on feedback using WBAs.

A key skill for successful clinician educators is the effective dissemination of scholarly innovations and research. Although there are many ways to disseminate scholarship, the most accepted and rewarded form of educational scholarship is publication in peer-reviewed journals.
This paper provides direction for emergency medicine (EM) educators interested in publishing their scholarship via traditional peer-reviewed avenues. It builds upon four literature reviews that aggregated recommendations for writing and publishing high-quality quantitative and qualitative research, innovations, and reviews. Based on the findings from these literature reviews, the recommendations were prioritized for importance and relevance to novice clinician educators by a broad community of medical educators.
The top items from the expert vetting process were presented to the 2016 Canadian Association of Emergency Physicians (CAEP) Academic Symposium Consensus Conference on Education Scholarship. This community of EM educators identified the highest yield recommendations for junior medical education scholars. This manuscript elaborates upon the top recommendations identified through this consensus-building process.

Background
Quantitative research is one of the many research methods used to help educators advance their understanding of questions in medical education. However, little research has been done on how to succeed in publishing in this area.
ObjectiveWe conducted a scoping review to identify key recommendations and reporting guidelines for quantitative educational research and scholarship.
MethodsMedline, ERIC, and Google Scholar were searched for English-language articles published between 2006 and January 2016 using the search terms, “research design,” “quantitative,” “quantitative methods,” and “medical education.” A hand search was completed for additional references during the full-text review. Titles/abstracts were reviewed by two authors (BT, PC) and included if they focused on quantitative research in medical education and outlined reporting guidelines, or provided recommendations on conducting quantitative research. One hundred articles were reviewed in parallel with the first 30 used for calibration and the subsequent 70 to calculate Cohen’s kappa coefficient. Two reviewers (BT, PC) conducted a full text review and extracted recommendations and reporting guidelines. A simple thematic analysis summarized the extracted recommendations.
ResultsSixty-one articles were reviewed in full, and 157 recommendations were extracted. The thematic analysis identified 86 items, 14 categories, and 3 themes. Fourteen quality evaluation tools and reporting guidelines were found.
DiscussionThis paper provides guidance for junior researchers in the form of key quality markers and reporting guidelines. We hope that quantitative researchers in medical education will be informed by the results and that further work will be done to refine the list of recommendations.
The nature and extent of terrestrial protected area coverage on the UK's Overseas Territories

Signatories to the Convention on Biological Diversity (CBD) agreed to the effective protection of at least 17% of the terrestrial environment by 2020 (Aichi Target 11). Here, we assess the coverage of terrestrial protected areas (land protected by legislation) on the UK's Overseas Territories. These 14 Territories are under the sovereignty of the UK, a signatory of the CBD, and are particularly biodiverse. Eight Territories have protected areas covering 17% or more of their land, but the extent of protection across these Territories as a whole is low, with only 4.8% of this land designated as protected. This protection covered 51% of sites already identified as of conservation importance (Important Bird and Biodiversity Areas), although only 8% of the area of these sites was protected. The expansion of effective protection to meet the 17% target provides an opportunity to capture the most important sites for conservation. Locally led designation will require an improvement in knowledge of the distribution and density of species. This, together with measures to ensure that the protection is enforced and effective, will require provision of resources. This should be seen as an investment in the UK meeting its obligations to Aichi Target 11.

Whispering gallery modes (WGMs) have been widely studied over the past 20 years for various applications, including biological sensing. While the WGM-based sensing approaches reported in the literature have shown tremendous performance down to single molecule detection, at present such sensing technologies are not yet mature and still have significant practical constraints that limit their use in real-world applications. Our work has focused on developing a practical, yet effective, WGM-based sensing platform capable of being used as a dip sensor for in-vivo biosensing by combining WGM fluorescent microresonators with silica Microstructured Optical Fibers (MOFs).
We recently demonstrated that a suspended core MOF with a dye-doped polymer microresonator supporting WGMs positioned onto the tip of the fiber, can be used as a dip sensor. In this architecture the resonator is anchored to one of the MOF air holes, in contact with the fiber core, enabling a significant portion of the evanescent field from the fiber to overlap with the sphere and hence excite the fluorescent WGMs. This architecture allows for remote excitation and collection of the WGMs. The fiber also permits easy manipulation of the microresonator for dip sensing applications, and hence alleviates the need for a complex microfluidic interface. More importantly, it allows for an increase in both the excitation and collection efficiency compared to free space coupling, and also improves the Q factor.
In this paper we present our recent results on microstructured fiber tip WGM-based sensors and show that this sensing platform can be used in clinical diagnostics, for detecting various clinically relevant biomarkers in complex clinical samples.

The impacts of La Niña-induced drought on Indian Vulture Gyps indicus populations in Western Rajasthan

Previous research on the catastrophic decline of the Gyps species complex has identified diclofenac, a non-steroidal anti-inflammatory drug administered to livestock, as the primary cause. Large-scale climatic phenomena, such as ENSO-induced drought, however have not been examined. Based on time series analysis of annual count data, 1996–2005, we provide evidence that ENSO synchronised population dynamics throughout western Rajasthan. Here, we ask whether El Niño Southern Oscillation (ENSO) can also explain the population dynamics of the Critically Endangered Indian Vulture Gyps indicus. We attribute this impact largely to two La Niña events, including the major event spanning 1999. We also examine between-village variation in resident vulture populations. Our results suggest that in several villages, Indian Vulture populations may have been partially buffered from the negative effects of drought when compared to other villages in the study. Finally, we discuss potential causes of buffering in these villages.

Summary
Pausanias has long been essential reading for archaeology students and those interested in reconstructing the topography of ancient poleis. Yet - as anybody who has been frustrated by such infuriatingly vague directions as 'not far from' or 'a little further on' can testify - Pausanias is not overwhelmingly interested in offering his readers a detailed guided tour of sites to see. Rather, the monuments that are described are the repositories of local narratives, both factual and fictional, that constitute an important part of the cultural heritage of a Greece now enslaved to Rome. In the description of the Argive agora, for example, the reader is introduced in short order to the tomb where Danaus' daughter Hypermnestra and her husband Lynceus are buried (2.21.2), the tumulus where Perseus interred Medusa’s head (21.5) and the underground bronze chamber in which Acrisius incarcerated Danae (23.7). Each polis that Pausanias visits grounds its unique identity in the specific matrix of myths and memories that are conveyed through such visible monuments.
Myth was not, however, confined to affairs within the polis. From at least the fifth century, diplomatic relationships between poleis had been articulated through the vocabulary of kinship (syngeneia), often explained in terms of mythical connections between the two communities. Thus, at about the time when Pausanias was writing (probably in the 160s and 170s CE), an inscription was set up in the Argive agora celebrating the kinship between Argos and Cilician Aegeae and noting that it dated back to the time when Perseus, son of Danae, travelled to Cilicia in his hunt for the Gorgons.

Traditionally, treatments of the Archaic Greek world have been dominated by discussion of the polis - a term that is often loosely (but not entirely erroneously) translated as “city-state.” Victor Ehrenberg, who in many ways pioneered modern research into the origins of the polis, described it as the very “foundation and support of Greek culture”; more recently, Oswyn Murray has characterized it as “the dominant form of government in the Greek-speaking world for roughly a thousand years, enabling city dwellers to control directly all or much of their own government, and to feel a local loyalty to an extent which no modern society has achieved.” In reality, this emphasis on the polis has tended to obscure the fact that in numerous regions of Greece, especially in the north and west, it was not the exclusive or even dominant form of sociopolitical organization until fairly late in the Classical period. On the other hand, there is no denying that much of the literature that survives from the Archaic period betrays the perspective of poets for whom the polis constituted an important point of reference, and this should at least serve to justify continued interest in how and when this characteristically Greek institution arose. To answer these questions, however, we first need to define what the polis was. Although it would be a mistake to assume that every polis developed in the same way or as a result of the same factors, there are nevertheless certain shared defining characteristics that can be identified.
